There different types of wheel barrow tires that require different methods to fix them. The tube type you patch the tube. The tubeless type you can plug the hole or put a patch on the inside of the tire.

For the tubeless tire if the tire has come off the rim it can be hard to get it to seal at the rim in order to get some air pressure in the tire. One of the easiest ways to force the sides out to seal on the rim is to take a ratchet tie down strap and tie it around the outside of the tire. now you should be able to get some pressure inside to make te tire beads pop on to the rim.
